POSTECH 연구팀, 초고해상도 광음향 현미경 개발의 새 지평 열었다

25일 POSTECH(포항공과대학교, 총장 김무환)은IT융합·전자전기·기계공학과 김철홍 교수·IT융합공학과 박별리 박사·통합과정 한문규 씨 기계·화학공학과 노준석 교수·기계공학과 통합과정 김홍윤 씨 연구팀이 광섬유의 근접장으로 얻은 광음향 신호를 영상화하는 데 최초로 성공했다고 밝혔다.

 

POSTECH에 따르면 그동안 광음향 현미경을 사용할 땐 일반적으로 광원과 샘플 사이에 일정 거리를 두고 렌즈로 빛을 모으는데, 회절 한계로 인해 빛을 한 점에 모으기 어려웠다.

 

이에 연구팀은 회절 한계를 뛰어넘기 위해 지름이 수십 나노미터(nm, 1nm=10억분의 1m) 단위인 끝이 가늘어지는 광섬유를 사용해 광원과 샘플의 거리를 회절 현상이 나타나지 않는 근접장 범위(수십 nm)로 유지하는 현미경 시스템을 개발했다.

특히, 광섬유를 둘러싼 금속을 제거함으로써 최초로 광음향 현미경에 적용했다는 점에 주목할 만하다. 기존 끝이 가늘어지는 광섬유는 금속의 영향으로 정확한 광음향 신호를 얻을 수 없어 광음향 현미경에 활용되지 못했다.

연구 결과, 광섬유 끝에서 소멸파 형태의 빛이 발생했으며, 샘플이 1.0±0.3마이크로미터(μm, 1μm=100만분의 1m)의 해상도로 영상화됨을 확인했다. 이는 적혈구를 영상화할 수 있을 정도의 해상도다.

이 결과는 광섬유를 활용한 초고해상도 광음향 현미경 개발의 초석이 될 연구성과로, 향후 실혈관 질환 및 암 등을 비롯한 다양한 질환의 근간이 되는 기초 생명 현상 연구기기로 활용이 기대된다.

한편, 국제 학술지 ‘레이저 앤 포토닉스 리뷰(Laser and Photonics Reviews)’에 최근 게재된 이 연구는 한국연구재단 중견기술, BRIDGE융합연구개발사업, 미래유망융합기술 파이오니어사업, 글로벌프런티어사업, 범부처전주기의료기기연구 개발사업, 산업혁신인재성장지원(R&D), BK21 사업의 지원을 받아 이뤄졌다.

According to POSTECH, when photoacoustic microscopes are used, light is generally collected with a lens at a certain distance between the light source and the sample, but it was difficult to collect the light to one point due to the diffraction limit.

 

In order to overcome the diffraction limit, the research team used a tapered optical fiber with a diameter of several tens of nanometers (nm, 1 nm = 1 billionth of a meter) to measure the distance between the light source and the sample in the near-field range where diffraction does not occur (tens of nm). developed a microscopy system to maintain it.

 

In particular, it is noteworthy that it was applied to photoacoustic microscopy for the first time by removing the metal surrounding the optical fiber. Conventional optical fibers with tapering ends could not be used for photoacoustic microscopy because accurate photoacoustic signals could not be obtained due to the influence of metal.

 

As a result of the study, it was confirmed that light in the form of evanescent wave was generated at the end of the optical fiber, and the sample was imaged with a resolution of 1.0±0.3 micrometers (μm, 1 μm = 1 millionth of a m). This is a resolution sufficient to image red blood cells.

 

This result is a research achievement that will serve as a cornerstone for the development of ultra-high-resolution photoacoustic microscopes using optical fibers, and is expected to be used as a research instrument for basic life phenomena that is the basis of various diseases, including true vascular disease and cancer.
